SARS-CoV-2 (COVID-19) Spike-Fc

The 2019 novel coronavirus (SARS-CoV-2) is a type of virus with a spherical shape, protrusions on the surface, and a crown-like virus when observed under an electron microscope. The virus gene is a continuous linear single-stranded RNA with a diameter of 75-160nm . Coronavirus particles are spherical or irregular in shape, have envelopes, and are 80-120 nm in size . Its genome 5 ' end with a cap structure, thereafter comprising 6-10 open reading frame ( the Open Reading Frames, the ORFs ). The first reading frame that occupies 2/3 of the genome encodes replicase, and the other 1/3 of the genome mainly encodes structural proteins, generally including spike protein ( spike, S ), envelope protein ( envelope, E ), and envelope protein ( membrance, M ), nucleocapsid ( N ). The E protein and M protein are mainly involved in the assembly process of the virus, and the N protein wraps the genome to form a nucleoprotein complex.

Among them, S protein is the most important surface membrane protein of coronavirus, which infects human respiratory epithelial cells by binding to human ACE2. The protein is a type I transmembrane protein, which contains two subunits , S1 and S2 . Among them, S1 mainly contains the receptor binding domain ( RBD ), and the S2 subunit can fix the entire S protein on the cell membrane and mediate virus-host cell and virus-cell membrane fusion. The S protein undertakes the functions of virus and host cell membrane receptor binding and membrane fusion. It is an important site for host neutralizing antibodies and a key target for vaccine design.

Illustration: Known HepG2 cell high expression of ACE2 protein (of PUBMED, the HPA) , the immune cells do not express ACE2 . In HepG2 and blank T cells were incubated 1.5ug of CoV-2 Spike-Fc protein, a secondary antibody incubation PE-anti human IgG for flow cytometry. The results showed that CoV-2 Spike-Fc protein can bind to HepG2 cells, but not ACE2- negative T cells; indicating that the purified CoV-2 Spike-Fc protein has biological activity.
